CFSE (Carboxyfluorescein Succinimidyl Ester) is a fluorescent cell staining dye used to monitor cell proliferation, division, and migration. As cells divide, CFSE fluorescence is equally distributed between daughter cells, allowing precise tracking of multiple generations by flow cytometry.
During cell proliferation, the fluorescent signal is distributed equally between daughter cells, causing fluorescence intensity to decrease by approximately half with each division. This predictable dye dilution allows researchers to quantify cell proliferation, identify cell generations, and evaluate immune responses using flow cytometry.
CFSE is extensively used in immunology, cancer biology, vaccine research, stem cell studies, and adoptive cell therapy development. It remains one of the most widely cited and trusted reagents for monitoring T-cell, B-cell, NK-cell, and stem cell proliferation.
